South Brunswick Township was established by an act of the State Legislature in 1798. The township began as a rural community with small settlements along major transportation routes. Early settlers took advantage of the fertile soil and favorable growing conditions in the area.

The township maintained its rural character throughout the 19th century, with increases in commercial and residential development occurring alongside new transportation routes. The Straight Turnpike (now Route 1) was built in 1804. The Legislature reduced the size of South Brunswick multiple times - first in 1872 with the creation of Cranbury Township, then adjusting Cranbury's boundaries in 1885, and finally with the formation of Plainsboro Township in 1919.

Kingston became prominent due to its location on the Lenape Assunpink Trail where it crossed the Millstone River. It was the most active town, situated on both the King's Road and Millstone River, supporting mills and taverns. Dayton was originally known as The Cross Roads, named after William L. Dayton who helped resolve railroad disputes.

The 20th century brought technological changes to South Brunswick. The New Brunswick and Trenton Fast Line began operation in 1900, and the New Jersey Turnpike opened in 1951. Kendall Park residential development started in the 1950s. The township population grew from about 18,000 in 1980 to over 33,400 today.

Despite rapid growth, much of the township's 42 square miles remain undeveloped with significant wetlands, forests and open space. Industry and commerce are well developed, with warehousing and distribution centers along the NJ Turnpike corridor and research facilities throughout the area.
